Il a \u00e9t\u00e9 largement d\u00e9montr\u00e9 que des kinases telles que c-Kit, Lyn et PDGFR, toutes des cibles cl\u00e9s du masitinib, sont fortement impliqu\u00e9es dans l\u2019activation des cellules inflammatoires et le remodelage des tissus fibreux.<\/p>\n<p>Il a \u00e9t\u00e9 d\u00e9montr\u00e9 que les mastocytes jouent un r\u00f4le cl\u00e9 dans l\u2019organisation et l\u2019activation de la cascade inflammatoire, non seulement en participant directement \u00e0 la destruction des tissus, mais aussi en recrutant et en activant d\u2019autres cellules immunitaires telles que les macrophages et les lymphocytes. Les kinases de c-Kit, Lyn et Fyn sont des kinases critiques pour le fonctionnement des mastocytes. Par cons\u00e9quent, les agents th\u00e9rapeutiques capables d\u2019inhiber ces kinases peuvent moduler l\u2019activit\u00e9 des mastocytes, entra\u00eenant par la suite une r\u00e9duction de l\u2019inflammation des tissus et donc des b\u00e9n\u00e9fices cliniques.\u00a0<em>In-vitro<\/em>\u00a0et les \u00e9tudes sur les animaux ont maintenant largement d\u00e9montr\u00e9 que le masitinib est un puissant inhibiteur de ces kinases, ce qui en fait un candidat m\u00e9dicament prometteur dans les maladies o\u00f9 l\u2019inflammation chronique joue un r\u00f4le central.[\/vc_column_text][\/vc_column_inner][\/vc_row_inner]<div class=\"dt-fancy-separator title-left h3-size title-color\" style=\"width: 100%;\"><div class=\"dt-fancy-title\"><span class=\"separator-holder separator-left\" style=\"border-color: #00477a;\"><\/span>M\u00e9canisme d&#039;action du masitinib dans la mastocytose syst\u00e9mique et dans le syndrome d&#039;activation des mastocytes<span class=\"separator-holder separator-right\" style=\"border-color: #00477a;\"><\/span><\/div><\/div><div class=\"ult-spacer spacer-69dcbda9c1f69\" data-id=\"69dcbda9c1f69\" data-height=\"20\" data-height-mobile=\"10\" data-height-tab=\"20\" data-height-tab-portrait=\"20\" data-height-mobile-landscape=\"10\" style=\"clear:both;display:block;\"><\/div>[vc_column_text]La mastocytose syst\u00e9mique indolente (MSI) est une maladie h\u00e9matologique caract\u00e9ris\u00e9e par un nombre anormal et une activation des mastocytes dans la moelle osseuse et d\u2019autres organes. La maladie est caract\u00e9ris\u00e9e par de multiples sympt\u00f4mes qui sont invalidants et peuvent, dans certains cas, mettre la vie en danger. Les sympt\u00f4mes associ\u00e9s \u00e0 la MSI sont principalement associ\u00e9s \u00e0 des troubles neurologiques (d\u00e9pression, fatigue, d\u00e9ficience cognitive, maux de t\u00eate), \u00e0 des troubles cutan\u00e9s (prurit, l\u00e9sions cutan\u00e9es), \u00e0 des bouff\u00e9es de chaleur et \u00e0 des troubles gastro-intestinaux.<\/p>\n<p>Les propri\u00e9t\u00e9s anti-mastocytes du masitinib semblent particuli\u00e8rement bien adapt\u00e9es au traitement de la mastocytose syst\u00e9mique indolente. Une r\u00e9duction de l\u2019activit\u00e9 des mastocytes est g\u00e9n\u00e9r\u00e9e par son action inhibitrice sur les tyrosines kinases sauvages de type c-Kit, Lyn et Fyn.
